The MouseTRAP™ test is a solid phase immunofi xed enzyme activity assay for the determination of osteoclastderived tartrate-resistant acid phosphatase form 5b (TRACP 5b) in mouse serum samples .
Summary and Explanation
TRAP is expressed in bone-resorbing osteoclasts and secreted into the circulation. Thus, serum TRAP has been considered as a potentially useful marker of bone resorption rate. However, several problems have prevented the development of specifi c TRAP assays including the presence of other interfering acid phosphatases in the blood circulation.
MouseTRAP™ Assay is specifi c for circulating TRACP 5b derived from mouse osteoclasts. And even more,MouseTRAP™ Assay detects ONLY the active TRACP molecules that are freshly liberated into the circulation.It does NOT detect old circulating inactive TRACP molecules or fragments. Thus, MouseTRAP Assay gives an accurate estimation of the amount of bone resorption rate at the time of sample collection.
Method Description
MouseTRAP™ Assay uses a polyclonal antibody prepared using generated recombinant mouse TRACP (2) as antigen.
In the test, the antibody is incubated in anti-rabbit IgG-coated microtiter wells. After washing, standard, control, and serum samples are incubated in the wells, and bound TRACP 5b activity is determined with a chromogenic substrate
to develop color. The reaction is stopped, and the absorbance of the reaction mixture is read in a microtiter plate reader, color intensity being directly proportional to the amount and activity of TRACP 5b present in the sample.
